چکیده

(1) Background: Congenital factor (F) VII deficiency is caused by mutations in the F7 gene. Patients with modest differences FVII levels may display large clinical severity. The variant p.A354V-p.P464Hfs associated reduced antigen and activity. aim of study was to investigate manifestation this underlying molecular mechanisms. (2) Methods: Analyses were conducted 37 homozygous patients. recombinant produced mammalian cells. (3) Results: We report a variation phenotypes, which points out genetic acquired components beyond as source variability. In contrast, patients displayed similarly plasma higher than its Comparative analysis samples from subset indicated presence an elongated indistinguishable migration. Treatment cells chemical chaperone 4-phenylbutyrate (4-PBA) improved intracellular trafficking increased secretion conditioned medium up 2-fold. However, effect 4-PBA on biological activity marginal. (4) Conclusions: Chemical chaperones can be used biochemical tools fate trafficking-defective variant.

Chemical chaperones assist intracellular folding to buffer mutational variations

Hidden genetic variations have the potential to lead to the evolution of new traits. Molecular chaperones, which assist protein folding, may conceal genetic variations in protein-coding regions. Here we investigate whether the chemical milieu of cells has the potential to alleviate intracellular protein folding, a possibility that could implicate osmolytes in concealing genetic variations. We f...

Factor VII Gene Defects: Review of Functional Studies and Their Clinical Implications

Coagulation factors belong to a family of plasma glycosylated proteins that should be activated for appropriate blood coagulation. Congenital deficiencies of these factors cause inheritable hemorrhagic diseases. Factor VII (FVII) deficiency is a rare bleeding disorder with variable clinical symptoms. Various mutations have been identified throughout the F7 gene and can affect all the protein do...

Some Biochemical and Electrophoretic Studies on Purified Prothrombin , Factor VII (

S TUDIES of the chemical nature of prothrombin have indicated a glycoprotein structure.2 Data obtained on purified bovine prothrombin have shown the reducing sugar content to be approximately 5 per cent, based on a reference standard of galactose and mannose.35 Laki et al.5 reported a value of 6.5 per cent in terms of glucose, and 1.6 to 1.7 per cent hexosamine. Resistance to thyroid hormone is an uncommon condition. We report the clinical and laboratory findings of a case with both resistance to thyroid hormone and, a reduced capacity to produce and respond to thyroid hormone. RTH is a disorder characterized by elevated circulating thyroid hormones, state of non-suppressed pituitary TSH secretion and refractoriness to hormone action in peripheral tiss...
